Proc. N atl. Acad. Sci. S. 54, 75 {1965). 2* Penicillins and Cephalosporins II. In the Host Carl W. Godzeski and Edwin H. Flynn Penicillin-Host Interactions The host environment is so different and complex compared to in vitro systems that to expect the mechanism of action of this or any other antibiotic agent to be sequentially identical in both environments is unreasonable. The relative importance of primary, secondary, or later effects as measured in the laboratory test tube may be markedly shifted by interactions of the host polyphasic system.

Methicillin action in vivo has been noted to be unexpectedly ineffective in a penicillin-resistant staphylococcal infection {DINEEN and MANNIX, 1962). That the apparent decreased potency may have a rational explanation was commented on by FROST and VALIANT {1964). These authors found that the route of infection and treatment were equally important in determining the outcome of the infection, using penicillinase producing staphylococci. Three days were required before in vivo penicillinase production took place.
